I am interested in various aspects of HIV-1 interactions with dendritic cells. Dendritic cells play a fundamental role in driving an appropriate immune response. Interestingly, HIV-1 is very inefficient at infecting dendritic cells. It has been shown that this restriction to HIV-1 can be alleviated by Vpx, a protein absent in HIV-1 but found in the related SIVmac. I am developing genetic approaches to identify the mechanisms by which HIV is restricted in dendritic cells. I am also interested in the innate response to HIV-1. The innate reponses to many pathogens and chemical agents have been identified and are mediated by families of pattern-recognition receptors (PRRs) including TLR, NLR and RLH. However, the innate sensors to HIV-1 remain elusive. I am developing various molecular approaches to identify the innate sensors to HIV-1. Importantly, the ability to manipulate these mechanisms will likely be important to develop better HIV-1 vaccines.
