This example is a 10-repeat unit with an Ala substitution near the middle, which formed single crystals. The backbone is in a quite extended conformation called "polyproline II", with phi,psi angles near -70,160 (use the measure tool to confirm that). The 3-fold sequence repeat is echoed in an approximate 3-fold structural repeat, with the Gly residues toward the center where their small size lets the 3 chains come close enough together to allow main-chain H-bonding (from a Gly NH of one chain to a Pro CO of a neighboring chain), as shown in purple. Try View2 for an end view. Choose View3 to see how the Ala substitutions (gold) are accommodated at Gly positions - the chains must move apart somewhat, and there are now H-bonds between chains through internal water molecules (as shown schematically in Fig. 14.3b). At all positions, the triple helix is also stabilized by external networks of water H-bonds (turn on "outer water" and "outer Hbonds", in View1), which connect backbone CO and hyPro OH groups, as also shown in Fig. 14.4. Choose View4 for an overview of this short section of collagen fiber (as in Fig. 14.2); an entire collagen fiber would be about 30 times this length. *{Kinemage 2}* shows a coiled-coil fiber formed of two long alpha helices spiralling around each other. This example is from a low-resolution crystal structure of tropomyosin, but it is very similar to the coiled-coils in intermediate filaments (Fig. 14.6b), to the tail section of myosin (Fig. 14.14), or to the leucine zipper dimerization motifs described in Chapter 10. The alpha-helices are righthanded, but the coiled-coil has a lefthanded supercoil. The sequence has a 7-residue repeat, with hydrophobic residues at positions a and d where they form the helix-helix contact. *{Kinemage 3}* shows the structure of myosin subfragment 1, which contains the actin-binding myosin "head" and a short part of the helical tail. The structure initially shown (as in Fig. 14.15) is the apo myosin S1 from chicken, solved at 2.8A resolution, with the two light chains in pink and yellowtint, and the heavy chain color-coded by domain. At the top is the site of binding to the actin fiber (thin filament of muscle), while the long helix at bottom continues on into a very long helical coiled-coil with a second myosin molecule, contributing to the thick filament. Animate, to see an approximation to the swinging motion of the myosin head that provides the working stroke for muscle contraction, moving the thick and thin filaments past each other. The second structure was constructed from the ADP/vanadate complex of a smaller fragment from Dictostelium myosin which shows the working conformational change, with the S1 tail portion superimposed onto its C-terminal end. The ATP-binding site is near the middle of the heavy-chain head region, shown in close-up in View3. Animate to see the local changes at that site, which are analogous to the switch II regions in G proteins, discussed in Chapter 13.
