Taxi (Yellow Taxi only) is best with a a $20-25 fare to the Medical Center (you must also pay any bridge or tunnel tolls).
But, by car:
Take the Grand Central Expressway (278) to the Triboro Bridge
and come south on FDR Drive to 34th St. Then go over one block to
First Ave and NYU Medical Center is at 540 First Ave (between 30th and
33rd Ave). There are at least 2 parking lots between 30th and 33rd
Aves, one on each side of the street.
By car:
Follow the New Jersey Turnpike to the Lincoln Tunnel and come across
Manhattan on whatever street you find yourself on (e.g., 38th or 40th or
42nd). Then go south on Second Ave to 30th St, turn left and then
left again on First Ave and you can find the above parking lots.
Taxi (Yellow taxi only) has $30 fixed fee from airport to anywhere in Manhattan.
Subway is quite reliable and perhaps best during rush hour. Certainly cheapest. Take the the free shuttle bus that visits all the airport terminals to the "A" train, which comes into Manhattan. You will have to change to the "4" or "5" train at Broadway-Nassau St to come up the East Side. NYU Medical Center is best served by 33rd St. stop, which requires another change to the local train ("6") at either Canal or 14th St. (just across the platform).
By car:
Take VanWyck to Long Island Expressway (West) to Midtown tunnel.
Go left out of tunnel following Downtown signs. Turn left again and
you will be at 2nd Ave and ~38th St. Turn right on 2nd Ave and then
left on 30th St and then left on 1st Ave.
Bus M16 runs up 8th Ave in front of Port Authority, across 43rd St and down 9th Ave behind Port Authority. Take Bus M16, which then goes across town along 34th St. Get off at 2nd Ave and 34th St and walk across to 1st Ave. For bus map, see MTA web site.
From Penn Station:
Get M16 bus along 34th St.
From Grand Central Station:
Walk.
NYU Medical Center is between 30th and 33rd Street on First Ave.
Go in the main door and look for the Skirball Instititute to your left
of the lobby (any guard can be asked for directions and there are usually
plenty of them around). I can be found on the 3rd floor, lab 13.
After exiting the elevators, turn away from the circular staircase and
enter the long hall where the labs are located, or you can go to the administrative
area next to the circular staircase and ask for directions.
